What are a blood substitute and a hemoglobin-based oxygen carrier (HBOC)? What is the state of the art in the development of  HBOCs? Which are the unsolved issues that up to now have hampered the FDA approval of a safe HBOC?  Is this situation dependent on the lack of knowledge in NO and oxygen physiology and biochemistry?  Is it better to produce a HBOC from outdated blood or from recombinant hemoglobin? Is there even a real need for HBOCs as therapeutic agents? In which clinical settings are HBOCs most needed? In which clinical situations is a HBOC a better therapeutic agent than donor blood?  These are just a few of the main questions that this special issue of /BBA Proteins and Proteomics/ on Hemoglobin-based Oxygen Carriers addresses thanks to contributions from key researchers in this field.

 

A. Mozzarelli

Andrea Mozzarelli is Full Professor of Biochemistry at the Faculty of Pharmacy, University of Parma, Parma, Italy.

The main research interest is the elucidation of structure-dynamics-function-regulation relationships of hemoglobins, pyridoxal 5-phosphate-dependent enzymes and green fluorescent proteins. The studies on hemoglobin has generated activities towards the development of a hemoglobin-based blood substitute. Protein immobilization via crystallization or encapsulation in silica gels has been exploited as a novel tool to select and isolate distinct tertiary and quaternary conformations, analyzed by spectroscopic and microspectroscopic methods. Advanced computational methods are applied to predict free energy of molecular interactions and to design protein ligands. He was the co-chairman of the conference on “International Visions on Blood Substitutes”, September 2006, Parma, Italy, within the frame of a European-funded project. He is the President of the XII International Symposium on Blood Substitutes that will be held in Parma, Italy, on August 25-28, 2009. He is member of the Editorial Board of BBA Protein and Proteomics, Current Medicinal Chemistry, and the 6th and 7th edition of Burger’s Medicinal Chemistry.
